A Phase 1 Study of CG0070 in Intermediate Risk Non-Muscle Invasive Bladder Cancer

Summary:

Background 

Intermediate-risk non-muscle invasive bladder cancer (IR NMIBC) is a form of bladder cancer that usually doesn’t spread deeply into the bladder wall or become more aggressive. However, it tends to come back often, even after treatment. Because of this, patients must go through lifelong checkups, which include uncomfortable procedures like cystoscopies and repeated bladder surgeries. Unfortunately, there are few effective treatments to stop the cancer from returning. 

What the Study Proposes to Address 

This study will test a new drug called CG0070, which is an oncolytic virus. That means it is a special type of virus designed to attack and kill cancer cells while helping the immune system recognize and destroy them. In this study, patients will first have most of their bladder tumors removed through surgery, leaving a small portion behind to help measure how well the treatment works. The drug will then be placed directly into the bladder once a week for six weeks, similar to how BCG treatments are given. After the first round of treatments, doctors will take a biopsy to see if the drug worked. Patients who respond well will continue with maintenance treatments every few months for a year. Researchers will also collect samples of tumor tissue, blood, and urine to look for specific biological markers that might predict how well the treatment works. In addition, they will study how the immune system reacts to the virus. 

Why This Research is Important 

If successful, this research could lead to a new and more effective treatment for patients with intermediate-risk bladder cancer. It could help reduce how often the cancer comes back and decrease the need for repeated surgeries and lifelong monitoring. By identifying biological markers that predict which patients respond best, doctors could personalize treatments and improve results. The study also aims to better understand how the immune system can be trained to fight bladder cancer, possibly leading to long-lasting protection against recurrence.
